Partnering Process


5 Steps

  1. Submit a non-confidential scientific overview with sufficient data to allow a preliminary scientific review. 
     Information Sheet
  2. Opportunity is reviewed and, upon acceptance, a Material Transfer Agreement (MTA) is drafted.
  3. MTA agreement is executed.
  4. Sirna ships chemically modified siRNAs.
  5. If applicable, results are reviewed and if results meet the performance criteria (see below) and if both parties are interested, a more formal evaluation can be established.

Our partnership process is well established and efficient, with an expert team focused on the scientific aspects of each opportunity. Our External Research Team mobilizes Senior Management from the appropriate disciplines to review each opportunity.

Of particular interest to Sirna is evaluation of technology platforms for drug delivery and oligonucleotide optimization, as well as, research collaborations exploring novel approaches in RNAi technology.

Sirna will distribute, free of charge, chemically modified siRNA to validate all opportunities that are accepted.


Delivery Vehicle Performance Criteria

Physical and Chemical Properties of Drug Delivery System
  • Reproducible assembly of functional complexes at 10mg scale
  • Assemblies less than 200nm diameter
  • Assembly zeta potential and polydispersity
  • siRNA encapsulation efficiency
  • Total siRNA in the assembly
  • No aggregation in 50% mouse/human serum
  • Chemical stability data of the assembly for >30 days
Activity in Cell-Based Assays
  • Greater than 50% reduction in SSB mRNA levels at siRNA concentrations <10 nM
  •  >5-fold window between SSB gene silencing IC50 and IC50 for reduction in viability
  •  Activity in at least 3 cell lines including HeLa, HepG2, Raw 264.7
In Vivo Performance in Mouse Models
  • Single i.v. dose at 1, 3 and 9 mg/kg
    • >50% reduction in SSB mRNA levels in target tissue at 1 mg/kg dose
    • <10-fold increases in ALT and AST at 9 mg/kg
    • No effect on body weight (48 hr)
